Abstract Title: Changes of mRNA expression for GABAA receptor in spinal cord on the alalgesic effect of subarachnoid transplantation of APA microcapsulized BCCs

ABSTRACT BODY

Introduction: To study the changes of mRNA expression for GABAA receptor2 and2 subunit in spinal cord on the alalgesic effect of subarachnoid transplantation of APA microcapsulized BCCs.
Case Report:Methods: SD rats were randomly divided into four groups ,each has five. Normal rats were used as control group (group C). Rats that right sciatic nerve was ligated were used as CCI group . Five to six hundred empty APA microcapsules (group PN) or 5~106 APA microcapsulized BCCs ( group BCC) were grated into subarachnoid space of CCI rats one week after operation. Allodynia and hyperalgesia were measured by Von- Frey filaments and CO2 laser dividually one week after transplantation. Segment four and five of lumbar spinal cord was taken out and mRNA expression for GABAA receptor2 and2 subunit was measured by RT-PCR. Results: Mechanical allodynia and heat hyperalgesia were observed in group CCI and group PN,but were alleviated in group BCC. The expression of mRNA for GABAA receptor2 and2 subunit in the spinal cord of group CCI and group PN was lower than that of group C and group BCC.There was no difference in the expression of mRNA for GABAA receptor2 and2 subunit between group BCC and group C.

Discussion: mRNA expression for GABAA receptor2 and2 subunit in the spinal cord of CCI rats was down-regulated. APA microcapsulized BCCs grafting can reverse the down-regulation of mRNA expression for GABAA receptor2 and2 subunit in the spinal cord of CCI rats. Recovery of GABAA-nergic neuron activity contributes to the analgesic effect of subarachnoid transplantation of APA microcapsulized BCCs.
